The depression is a multifacetic disorder that can display an ample variety of symptoms between which are psychomotor alterations, diminution of libido etc. Patients with depression exists sorne signs that are marking biological for the diagnosis of the depression as the increase in the plasma levels of cortisol. On the other hand, postnatal administration of clomipramine (CLI) to rats male produces behavioral abnormalities during adulthood which closely resemble sorne of the symptoms observed in human depression. Therefore, this procedure has been suggested as an animal model of depression. Exist few evidences about the activity and sensitivity of the hypotalamic-pituitary-adrenal axis (HPA) in rats CLI. The objectives ofthe present study were: to determine the response of axis HPA, through the evaluation of corticosterona (C) prior to different stimuli like the application of a estresor, as acute as chronic, prior to the execution of the masculine sexual behavior (MSB) and prior to the administration of cholinergic agonists as oxotremorine (OXO) and physostigmine (PHY) in the rats treated postnatally with CLI. In addition, to evaluate the circadian activity of C and finally cholinergic sensitivity in this animal model of depression. Male pups rats Wistar were injected se twice a day from 8 to 21 of age. Each litter received the same treatment. One group received CLI (15 mg/kg) in each injection, while Control (CON) group received saline solution. In adulthood, ali subjects were submitted to the forced swim test and motor activity to evaluate to effect of postnatal clomipramine treatment. Rats CLI and CON were randomly assigned to one of following experiment: l) Validity the animal model of depression trough to evaluate the circadian activity of C-; the measurements were made at the beginning and the middle of the phases of light and the dark. 2) Activation of HPA axis by means of stress by cold water immersion (IMS) as much of acute way (l day) like chronic (10 days) in both phases of the cycle of light-darkness. 3) Activation of HPA axis by means of the administration of cholinergic drug as PHY (0.4 mg/kg ip) and OXO (0.4, 0.8 mg/kg ip), in both phases of the cycle of light-darkness. 4) Activation of HPA axis to trough of the MSB; previously were made four tests of spontaneous MSB. After the last test, the animals immediately were sacrificed to evaluate the plasma levels of C. 5) Evaluation of cholinergic sensitivity by means of the tests of Porsolt, motor activity, and rectal temperature prior to the administration of OXO to different doses (0.2, 0.4, 0.8, l.6 mg/kg, ip). The postnatal treatment with CLI increased per se the immobility time in the test of Porsolt. In addition to alter the sexual activity, decreasing the percentage of subjects to display mounts, intromissions, ejaculations and to alter the different parameters copulatories. l) The rats treated with CLI showed alterations in the circadic activity of C, with an increase in the levels of C in the phase of light. 2) The response to stress by IMS was dependent of the duration, as well as of time of the exhibition. Whereas acute stress elevated the levels of C in both phases of the cycle of the light-dark in both groups (CON and CLI), chronic stress by IMS only increased the levels of C in the phase of light in the rats treated with CLI. 3) The administration of PHY increased the plasma levels of C only in the phase of light in the rats treated with CLI. This same response it was observed with the administration of OXO with the dose of 0.4, whereas the dose of 0.8 increased the levels of C in both phases of the cycle of light-darkness of way similar to the group contr<?l. 4) The activation of HPA axis to the exhibition to CSM increasing the levels of C in both groups. Nevertheless this increase is smaller in the rats treated with CLI respecting the rats CON after of the exposure MSB. 5) lt was not observed cholinergic hypersensitivity in the rats treated postnatally with CLI in the different behavior to evaluate, with the exception of the rectal temperature. The results shows that the postnatal treatment with CLI increases the time of immobility in the test of Porsolt, alters to the circadian activity of corticosterone and the response of HPA axis to different stimuli like the sexual behavior, stress, the administration of cholinergic drugs. Also, the administration of CLI does not induce cholinergic sensitivity.
