Por lo tanto, el desarrollo de películas y recubrimientos comestibles para los alimentos requiere de la incorporación de compuestos bioactivos para aumentar su funcionalidad y capacidad protectora. El aislado proteico de suero de leche (WPI) y el almidón nativo de papa (NPS) en combinación generarán películas con propiedades complementarias entre sí. Por otro lado, recursos naturales de bajo costo como los desechos agroindustriales, como las semillas del fruto de tamarindo, ofrecen una alternativa más en la obtención de compuestos bioactivos que pueden ser incorporados a las matrices de películas. En este trabajo se evaluó el uso de un extracto polifenólico de las semillas de tamarindo (TSE) en la formación de películas comestibles a partir del WPI y NPS, por lo que se determinaron los cambios en sus propiedades mecánicas, antioxidantes y antimicrobianas. Así como la estabilidad de sus propiedades en el almacenamiento y su aplicación bajo condiciones de laboratorio en carne de cerdo molida con la adición de 2% de NaCl. Lo anterior se realizó en tres etapas: 1) Optimización de las condiciones de elaboración de la matriz base para las películas de WPI-NPS mediante la metodología de superficies de respuesta, matriz en la que posteriormente se incorporó el TSE. Por lo que se evaluó la variación inducida sobre las propiedades mecánicas, color y antioxidantes durante el almacenamiento de las películas hasta por 35 días. 2) Evaluación de la actividad antimicrobiana de las películas WPI-NPS incorporadas con TSE sobre sepas microbianas de interés en el sector cárnico. La posible sinergia del TSE con otro antimicrobiano como la nisina también fue evaluada. 3) Aplicación de las películas WPI-NPS sobre carne de cerdo molida frente a un factor pro-oxidante como es el cloruro de sodio al 2%. Un sistema de evaluación a partir de cilindros se usó para aplicar las películas y determinar los cambios en el color, estabilidad oxidativa y antioxidante en la carne. Los resultados fueron los siguientes: 1) Para optimizar la producción de películas WPI-NPS se obtuvieron las siguientes condiciones de elaboración a partir de un diseño rotatorio central 2^3: 6.2 g/100 mL de glicerol en la solución formadora de películas (FFS), a pH de 7.9 y temperatura de desnaturalización proteica (PDT) de 76 °C, que predijeron las siguientes propiedades mecánicas: E (Módulo de Young’s) = 0.07 MPa,  (Fuerza a la tensión) = 1.94 MPa y %ɛ (Porcentaje de elongación) = 83.6 %, con una deseabilidad máxima del modelo de 0.86. La optimización fue constatada, produciendo películas control (sin la incorporación de TSE) y películas con dos diferentes niveles de TSE adicionado (1.26 y 12.6 mg/100 mL de FFS). Una vez secas las películas, se evaluaron nuevamente sus propiedades, encontrando una mejora (p < 0.0001) en las propiedades mecánicas al incrementar en 8% el %ɛ, y disminuir en 14% y 50% los parámetros de  y E, respectivamente. Las películas con TSE tuvieron una coloración rojiza persistente (p < 0.0007) durante el almacenamiento. Respecto a la actividad antioxidante inducida por el TSE, esta aumentó hasta 2.7 veces. Cabe señalar, que el TSE por sí solo, tuvo un contenido de polifenoles (187.0 ± 0.11 mg GAE/g) y actividad antioxidante (62.5 ± 0.04% para DPPH• y 45.6 ± 0.14% para ABTS+•) alta comparada con otras fuentes de antioxidantes. Todas las características de las películas adicionadas con TSE permanecieron estables, aún después de 35 días de almacenamiento. 2) Se determinó la actividad antimicrobiana por la técnica de difusión en pozos. El TSE tuvo actividad (p < 0.04) frente a E. coli, L. innocua y B. thermosphacta, cepas microbianas de relevancia por su relación con la salud del consumidor y resistencia al procesado de los productos cárnicos (temperaturas de refrigeración, sales y largos periodos de almacenamiento). Dicha actividad se redujo durante el proceso de formación de las películas (p < 0.02), específicamente en la FFS y después del secado en la fracción soluble de la película. Sin embargo, en la prueba para encontrar una posible sinergia del TSE con nisina, el resultado fue negativo, posiblemente, la actividad de la bacteriocina se redujo en presencia de los polifenoles del extracto. 3) Previamente se implementó un sistema de cilindros de PVC para colocar las películas u otro recubrimiento sobre el alimento, bajo condiciones de refrigeración (4 °C). Una funda de papel encerado (WPS) se usó como control en la evaluación. La carne de cerdo (animales de una misma genética y alimentación) con composición constante (4% de grasa) con 2% de NaCl añadido como factor pro-oxidante, se usó para evaluar la actividad protectora por el uso de las películas. Cambios en la apariencia y en la estabilidad oxidativa hasta por 14 días de almacenamiento fueron evaluados. Adicionalmente, se generó información de la posible liberación y difusividad del TSE hacia la carne, al analizar la respuesta a nivel superficial (primeros 5 mm de profundidad) e interna (entre 5 y 20 mm de profundidad) en el sistema cilíndrico con carne. Respecto al TSE, resultó ser un compuesto sin difusividad de la película hacia el alimento, actuando principalmente a nivel superficial por contacto induciendo una menor (P < 0.0001) oxidación lipídica y proteica; mientras que al interior de la carne se observó una mayor (P < 0.02) luminosidad y actividad antioxidante. En la comparación de las diferentes películas a los 14 días de almacenamiento, en la carne cubierta con las películas de 12.6 mg de TSE se redujo la luminosidad y los tonos amarillos (p < 0.003), mientras que los tonos rojos fueron constantes (p > 0.5). La actividad antioxidante por FRAP arrojó las mayores diferencias, manteniéndose constante cuando se usaron películas con 1.26 mg de TSE. La oxidación de la fracción proteica y lipídica a los 7 días de almacenamiento fue inversamente proporcional a la concentración de TSE usada en las películas. A los 14 días de almacenamiento, la oxidación en la carne protegida con películas al bajo nivel de TSE fue baja, contrario a lo observado con el nivel alto que incrementó la oxidación al actuar como pro-oxidante. En general, la elaboración de películas comestibles a partir de WPI y NPS incorporadas con un TSE, permitió la generación de películas estables, con propiedades mecánicas optimizadas, y actividad antioxidante y antimicrobiana extra, que lograron reducir los cambios oxidativos a nivel superficial en un alimento susceptible a la oxidación como es la carne, siendo 1.26 mg de TSE la concentración adecuada para aportar una protección extra a la carne de cerdo.; The growing demand of nutritive foods with a longer shelf life, has increased the development of new packages; but it is indispensable that biodegradables materials are include in order to decrease pollution. Therefore, the development of edible films and coats for food require the incorporation of bioactive compounds to enhance its´ functionality and protector capacity. The way protein isolate (WPI) in combination with native potato starch (NPS) will generate films with complementary properties inwardly. Otherwise, the low cost natural resources such as agroindustrial wastes, like tamarind seeds, offer other alternative for the obtaining of bioactive compounds which can be incorporate to the films matrixes. In this project changes in mechanical, antioxidants, and antimicrobial properties by the use of a polyphenolic tamarind seeds extract (TSE) in the elaboration of WPI-NPS edible films were evaluated. Also, the storage stability of properties and the application under laboratory conditions in ground meat added with 2% of NaCl were evaluated. All of the above, was done in three stages: 1) Elaboration conditions optimization of WPI-NPS films matrix through response surface methodology, after, in the same matrix the TSE was added. So that, the induced variation over the mechanical, color, and antioxidants properties was measured during 35 days of storage. 2) Evaluation of the antimicrobial activity in WPI-NPS films added with TSE against microbial strains of interest for meat industry. The possible synergy between TSE with nisin was also evaluated. 3) Application of WPI-NPS films over ground pork in the face of a pro-oxidant factor such as 2% of sodium chloride, in an evaluation system of cylinders was used to determine changes in the color, oxidation and antioxidant activity in the meat. The results were the following: 1) For the manufacturing of the WPI-NPS films a central rotatory design 2^3 was used, and the following conditions were obtain: 6.2 g/100 mL of glycerol in the film-forming solution (FFS), at pH 7.9, and a protein denaturalization temperature (PDT) of 76 °C, that predict the following mechanical properties: E (Youngs’ modulus) = 0.07 MPa,  (Tensile strength) = 1.94 MPa, and %ɛ (elongation percentage) = 83.6%, with a maximum model desirability of 0.86. The optimization with a control films and WPI-NPS edible films with two different levels of TSE (1.26 y 12.6 mg/100 ml of FFS) was verified. Once the films were dry, it´s properties were evaluated again, finding an improvement (p < 0.0001) in the mechanical properties by the increase of 8% of %ɛ, and the decrease of 50% and 14% of  and E, respectively. The TSE films had a persistence reddish coloration (p < 0.05) during storage. In regard to, the antioxidant activity induce by TSE was increased up to 2.7 times. It should be noted, that TSE by itself, had a high polyphenol content (187.0 ± 0.11 mg GAE/g) and an antioxidant activity (62.5 ± 0.04% for DPPH• and 45.6 ± 0.14% for ABTS+•) compared with other antioxidant sources. All the characteristics of films added with TSE stay stable, even after 35 days of storage. 2) The antimicrobial activity evaluated by the wells diffusion technic. The TSE had activity (p < 0.05) against E. coli, L. innocua and B. thermosphacta, these strains are relevant for the costumer health because their resistance to the processing of meat products (refrigeration temperatures, salts and a long storage periods). This activity decreased during the films formation process (p < 0.02), specifically in the FFS and after drying process, in the film soluble fraction. However, in the test made to find a possible synergy between TSE and nisin, the result was negative, possibly, the bacteriocin activity was reduced in presence of the extract polyphenols. 3) Previously was implemented a system of PVC cylinders to place the edible films or other coat in contact with meat, under refrigeration conditions (4 °C). A wax-paper sheath (WPS) was included as a control. The pork meat (from animals with the same genetic and feed) with a constant composition (4% fat), with 2% of NaCl added (as a pro-oxidant factor) was used to evaluate the protective activity by the used of the films. Appearance changes and oxidative stability over 14 days of storage were evaluated. Additionally, information about the possible release and diffusivity of TSE to the meat was generated by analyzing the response in a superficial level (first 5 mm depth) and internally (between 5 to 20 mm depth) in meat cylinders system. Regarding TSE, compound have not diffusivity from the film to the meat, acting mainly at the surface level by contact inducing a lower (p < 0.0001) lipid and protein oxidations; meanwhile in the interior of the meat a higher luminosity and antioxidant activity was measurement (p < 0.02). In the comparison of the different films at 14 days of storage, the films with 12.6 mg of TSE in the meat reduced (p < 0.003) the luminosity (L) and yellow tones (a*), but red tones were constant (p > 0.5). The antioxidant activity by FRAP showed major differences, staying constant when films with 1.26 mg TSE were used. The protein and lipid oxidation fraction, at the 7 days of storage was inversely proportional to the TSE concentration added in the films. At 14 days of storage, the oxidation remains low, only with the low TSE addition level, contrary to the high level that increase oxidation to act as a pro-oxidant. In general, the elaboration of WPI-NPS edible films added with TSE, stable films were generated with optimized mechanical properties, and extra antioxidant and microbial activity, which achieved to reduce oxidative changes at superficial level of food susceptible to oxidation just like meat, been 1.26 mg of TSE the adequate concentration to provide extra protection to the pork. %G spa %[ 2023-01-18 %9 info:eu-repo/semantics/doctoralThesis %~ UAM %W UAM
