Durante el sueño se ha mostrado que ocurren los mayores picos de liberación de diversas hormonas involucradas en la reproducción. En el sueño de ondas lentas ocurre la liberación de la Hormona Luteinizante (LH) y en el primer episodio de sueño MOR, se libera la Testosterona (T). Asimismo, se ha evidenciado que la supresión o reducción de sueño promueve la aparición de diferentes alteraciones reproductivas, tales como: cambios en la Conducta Sexual Masculina (CSM), daño en órganos andrógeno dependientes y cambios histomorfológicos del testículo y de la próstata ventral. Sin embargo, aún no ha sido descrito el efecto temporal de la restricción crónica de sueño MOR (RCSMOR), sobre la CSM, análisis del eyaculado e histomorfología de las vesículas seminales, así como durante el periodo de recuperación (REC). Por lo cual, el objetivo del presente trabajo fue evaluar el efecto de la RCSMOR sobre la CSM, calidad espermática e histomorfología de las vesículas seminales, durante la RCSMOR y en el periodo de REC. Se utilizaron 24 ratas macho, las cuales fueron colocadas de manera aleatoria en los siguientes grupos (n=4): 1) Control, 2) RCSMOR 7 días, 3) RCSMOR 14 días, 4) RCSMOR 21 días, 5) REC 7 días y 6) REC 14 días. La RCSMOR se realizó mediante la técnica de plataformas múltiples modificadas, los animales se mantuvieron con una vigilia prolongada de 20 h, permitiéndoles dormir sólo 4 h al día. Al final de la RCSMOR a los grupos 5 y 6 se les permitió recuperarse por 7 y 14 días, respectivamente. Se evaluó la CSM de todos los grupos, considerando los siguientes parámetros: latencias a monta, intromisión y eyaculación, número de montas e intromisiones, duración de la serie eyaculatoria e índice de intromisión. La obtención de muestras del eyaculado y de las vesículas seminales, se hicieron cada 7 días durante el periodo de RCSMOR y REC. El análisis del eyaculado se realizó el mismo día de las pruebas de CSM. Los órganos disectados fueros fijados en Bouin para su posterior procesamiento siguiendo la técnica histológica estándar para microscopia de luz, los cortes obtenidos (7µm) fueron teñidos con Hematoxilina-Eosina (H-E) para su análisis cualitativo y cuantitativo en el software Axio Vision Rel 4.8. Los resultados mostraron que la RCSMOR disminuyó la calidad del eyaculado desde la primera semana de RCSMOR, afectando la concentración, movilidad y viabilidad espermática, aunque no modificó la CSM. En cuanto a la morfología de la vesícula seminal, la RCSMOR a partir de los 7 días alteró la morfología del epitelio secretor, con una disminución de los pliegues sinuosos y de la secreción en criptas de la mucosa y lumen; así como el área total y altura del epitelio. Con estos resultados, concluimos que la RCSMOR alteró la histomorfología de las vesículas seminales produciendo una disminución de las secreciones que conllevaron a una reducción de la calidad y transporte espermático, aunado a la modificación del tapón seminal.; The sleep is a physiological process of vital importance for the integral health of the organisms and is fundamental for the optimal performance of the diverse daily activities. During the sleep it has been shown that occur the greatest peaks of release of various hormones involved in the reproduction. In slow waves sleep, occurs the release of Luteinizing Hormone (LH) and on the first episode of rapideyes movements sleep (REM), Testosterone (T) release. Likewise, it has been shown that suppression or reduction of sleep produces the appearance of different reproductive alterations, such as: changes in Male Sexual Behavior (MSB), damage in organs androgen-dependent and histomorphology changes of the testicle and in the ventral prostate. However, it has not been described the temporal effect of the chronic REM sleep restriction (REMSR), on the MSB, analysis of the ejaculate and histomorphology of the seminal vesicles, as well as during the recovery period (REC). Therefore, the aim of the work was to evaluate the effect of the REMSR on the MSB, the spermatic quality and histomorphology of the seminal vesicles, during the REMSR and in the REC period. Twenty-four male rats were used, which were randomly placed in the following groups (n = 4): 1) Control, 2) REMSR 7 days, 3) REMSR 14 days, 4) REMSR 21 days, 5) REC 7 days and 6) REC 14 days. The REMSR was performed by a modified multiple platform method, the animals were kept with a prolonged wake of 20 h, allowing them to sleep only 4 h per day. At the end of the REMSR, groups 5 and 6 recovered for 7 and 14 days, respectively. The MSB of all the groups was evaluated, considering the following parameters: mounts, intromission and ejaculation latencies, number of mounts and intromissions, duration of the ejaculatory series and copulation rate. Obtaining samples of the ejaculate and the seminal vesicles was done every 7 days during the period of REMSR and REC. The analysis of the ejaculate was done on the same day as the MSB tests. The organs dissected were fixed in Bouin for later processing them following the standard histological technique for light microscopy, the sections obtained (7 μm) were stained with Hematoxylin-Eosin (HE) for qualitative and quantitative analysis in the Axio Vision Rel software. 4.8. The results showed that the REMSR decreased the quality of the ejaculate from the first week of REMSR, affecting the concentration, motility and sperm viability, although it did not modify the MSB. Regarding the seminal vesicle morphology, the REMSR after 7 days altered the morphology of the secretory epithelium, with a decrease in the sinuous folds and secretion in the crypts of the mucosa and luminal region; as well as total area and height of the epithelium. With these results, we concluded that the REMSR altered the histomorphology of the seminal vesicles, producing a decrease in the secretions that led to a reduction in the quality and sperm transport, joined with the modification of the seminal plug. %G spa %[ 2023-01-18 %9 info:eu-repo/semantics/masterThesis %~ UAM %W UAM
