Entre los procesos involucrados en el desarrollo de hongos entomopatógenos sobre la cutícula de los insectos se encuentra la formación de hifas y otras estructuras especializadas que secretan enzimas degradadoras, como quitinasas, proteasas, lipasas y moléculas surfactantes, conocidas como hidrofobinas (St. Leger et al, 1998). Los hongos se caracterizan por la formación de biopelículas hidrofóbicas, siendo esta una característica importante para la biodegradación de hidrocarburos (Ortiz et al, 2003). Crespo et al, 2002 reportaron que usando n-hexadecano como substrato, la capacidad entomopatogénica de Beauveria bassiana incremento en el gorgojo del fríjol. Por otro lado Lecanicillium (anteriormente Verticillium) ha sido reportado como productor de quitinasas extracelulares (Matsumoto et al, 2004). El objetivo inicial de este trabajo fue la evaluación de la capacidad degradadora y el crecimiento de hongos entomopatógenos y micoparásitos en presencia de hidrocarburos; veintidós cepas de Lecanicillium y una de Beauveria bassiana fueron cultivadas en medio sumergido y superficial. Puro tolueno o n-hexano o la mezcla (en una relación de 17:83 porciento) fueron empleados, al mismo tiempo que extracto de levadura, glucosa o quitina coloidal. De acuerdo con los resultados, todas las cepas de Lecanicillium fueron capaces de crecer en medio con hidrocarburos con y sin la adición de otras fuentes de carbono (extracto de levadura, quitina coloidal o glucosa). Con la presencia de hidrocarburos se observó inhibición en el desarrollo del hongo, presentando un micelio fino, escaso, en aglomerados, con un número considerable de esporas sin germinar. El diámetro de las hifas fue significativamente menor con la presencia de n-hexano, indicando que la inhibición es mayor que con tolueno. Fue medido el consumo de n-hexano y tolueno en cultivos sumergidos con o sin quitina coloidal de las cepas EH-460, 157 y 2149 (que presentaron el crecimiento más alto), la degradación fue determinada de 41% a 57% después de las 400 h, observando mayor degradación con tolueno que con n-hexano. La cepa 157 presentó el consumo más alto (15.7 g tolueno/m3 ) y velocidad máxima de consumo (7.9X10-3 g tolueno/m3 •h). Cuando se añadió quitina al medio de cultivo con n-hexano esta fue consumida en las primeras horas, mientras que en medio con tolueno se observó un efecto diuxico. La actividad de endoquitinasas (Endo) y N-acetil-hexosaminidasa (NHasa) fue determinada de los medios con o sin quitina, por tanto estas enzimas también son producidas en presencia de hidrocarburos. El efecto del tolueno con la presencia de quitina fue medido en la cepa 2149. Estos experimentos confirman la expresión de NHasa y Endo (una actividad máxima de 0.27 mU/ml y 11 U/ml respectivamente) en medio sin quitina como inductor. Con la finalidad de analizar la expresión de la hidrofobina SC3 se extrajo ARN de L. lecanii 2149, crecido en medios de cultivo con glucosa o fructuosa, se realizó la retrotranscripción hacia ADNc y se amplificó la región específica de hidrofobina SC3 por la técnica de PCR, utilizando cebadores diseñados a partir de la secuencia del ADN complementario de la hidrofobina SC3 de Schizophillum commune, reportada en el Gene Bank.; The filamentous fungi are characterized by their capacity to colonize hydrophilic and hydrophobic substrates, such as the cuticle of insects. Among the processes involved in the development of entomopathogenic fungi on the cuticle of insects it is found the hyphae formation and other specialized structures that secrete degrading enzymes degradadoras, such as chitinases, proteases, lipases and surfactants molecules, known as hydrophobins (St. Leger et al, 1998). The fungi are characterized by the formation of hydrophobic biofilms being this an important characteristic for the biodegradation of hydrocarbons (Ortiz et al, 2003). Crespo et al, (2002) reported that using n-hexadecane as the substrate, the entomopathogenic capacity of Beauveria bassiana increased to bean weevil. On the other hand Lecanicillium (previously named Verticillium) has been reported to be an extracelullar chitinases producer (Matsumoto et al, 2004). The initial aim of this work was the evaluation of degrading capacities and growth of entomopathogenic and mycoparasitic fungi in presence of hydrocarbons; twenty two strains of Lecanicillium and one of Beauveria bassiana were cultivated in submerged and superficial media. Pure toluene or n-hexane or their mixtures (in a percentage ratio 17:83) were used, along with yeast extract, glucose or colloidal chitin. According to the results, all the strains of Lecanicillium were able to grow using hydrocarbons with or without the addition of other carbon sources. With the presence of hydrocarbons it was observed inhibition in the development of the mycelia presenting fine, scarce, in agglomerates with a considerable amount of non germinating spores. The hyphae diameter was significantly smaller with the presence of pure n-hexane, indicating that the inhibition is bigger than with pure toluene. Was measured the n-hexane and toluene consumptions in submerged cultures with or without colloidal chitin of the strains EH-460, 157 and 2149 (that presented the highest growth), degradation was determined as 41% to 57% after 400 h, and it was observed higher degradation with toluene than with the n-hexane. The strain 157 presented the highest consumption (15.4 g toluene/m3 ) with a maximum speed of consumption (7.9x10-3 g toluene/m3 *h). When adding chitin to the medium culture with n-hexane it was consumed in the first hours, while what in medium with toluene was observing a diauxic effect. The activity of endochitinases (Endo) and N-acetil hexosaminidase (NHasa) was determined from the media with or without chitin, therefore these enzymes were also induced by the presence of hydrocarbons. The effect of the toluene with presence of chitin was measured in the strain 2149. These experiments confirmed the expression of NHasa and Endo (a maximum activity of 0.27 mU/ml and 11 U/ml respectively) in media without chitin as inducer.
