Ex Willd) M. C. Johnst can be considered a potential hyperaccumulator of chromium (Cr) and lead (Pb). This paper presents a protocol for the establishment of Prosopis laevigata cell suspension culture with the aim of determining the effect of Cr(VI) and Pb(II) on the growth and accumulation in P. laevigata, well as, the competition of Cr(VI) and Pb(II) by Mn(II) and Zn(II) transporters, respectively. The callus used for this purpose were obtained from cotyledons of 15 days, cultured on Murashige and Skoog medium supplemented with 2,4- dichlorophenoxyacetic acid (2,4 -D, 1.5 mg L-1 ) and kinetin (KIN, 1.0 mg L-1 ). The growth kinetics of cultured cells in suspension in the absence of MP showed a specific growth rate (µ) of 0.0491d-1 and doubling time (td) of 14.1 d with a viability of 80 %. While the suspension cultures added of 1 mM Cr(VI) or Pb(II) showed a µ and a td of 0.0633 d-1 , d 10.94 for Cr(VI) and 0.0620 d -1 , 11.17 d for Pb(II). To determine the growth and accumulation of Cr and Pb bioassays were performed with different concentrations of Cr and Pb (0.0 – 3.0 mM) was observed in the cultured cells with 1.0 mM Cr(VI) relative growth (CR) was significantly higher (122 %) than the control, while the CR of the cultured cells with 2.0 and 3.0 mM of Cr(VI) was lower than in the control treatment (81 % and 58 % respectively). In the case of Pb was observed that cells cultured with 1 mM Pb(II) no significant differences in CR relative to the control, while those which were grown to 2.0 to 3.0 mM Pb CR was significantly less than control (81 % and 70% respectively). With respect to the accumulation of Cr and Pb by P. laevigata cell suspensions was observed that the cells accumulate more Pb (14,086-30,079 mg Pb Kg -1 dry weigth (DW)) to Cr (2379-7194 mg Kg -1 DW)). It was further observed that as the concentration is increased Cr or Pb in between both metal accumulation in biomass was greater. To determine the competence of the Cr(VI) by transporters Mn(II), a bioassay with Cr(VI)/ Mn(II) (0.0, 0.5 and 1.0 mM) was carried out on cell suspension cultures. Cells showed significant tolerance to growth of 83-98 % in all concentrations and combinations of Cr and Mn (0.0, 0.5, 1.0 mM). When the Cr(VI) with or without Mn(II) were added to the culture medium, the cells showed highest accumulation efficiency for non- essential (Cr) metal over essential (Mn) metal. Finally, to determine the competence of the Cr(VI) by transporters Mn(II), a bioassay with Pb(II)/Zn(II) (0.0, 0.5, and 1.0 mM) was carried out on cell suspension cultures. Cells showed significant tolerance to growth (CR>60%) at all concentrations and combinations of Pb(II) and Zn(II) (0.0, 0.5 and 1.0 mM). When the Pb(II) with or without Zn(II) were added to the culture medium, the cells showed the highest accumulation efficiency for non-essential (lead) metal over essential (zinc) metal (bioconcentration factor (FBC) values for Pb  FBC values for Zn; 2-33 times). Scanning electron micrographs evidenced the accumulation of Cr and Pb in the cells walls. These results provide insights about the tolerance and accumulation mechanisms of Cr and Pb occurring in P. laevigata.; Estudios en cultivos in vitro indican que Prosopis laevigata (Humb & Bonpl. Ex Willd) M. C. Johnst. puede ser considerada como una especie potencialmente hiperacumuladora de cromo (Cr) y plomo (Pb). En este trabajo se presenta un protocolo para el establecimiento de un cultivo de células en suspensión de P. laevigata, con el objetivo de evaluar el efecto del Cr(VI) y Pb(II) sobre el crecimiento, y su acumulación en los cultivos celulares, así como, la competencia del Cr(VI) y Pb(II) por los transportadores de Mn(II) y Zn(II), respectivamente. Los callos fueron inducidos a partir de cotiledones de 15 días de edad cultivados en medio Murashige y Skoog adicionado con 1.5 mg L-1 de ácido 2,4-diclorofenoxiacetico (2,4-D) y 1.0 mg L-1 de cinetina (KIN). La cinética de crecimiento, de las células en suspensión cultivadas en ausencia de metales pesados (MP), presentaron una velocidad específica de crecimiento (µ) de 0.0491d-1y tiempo de duplicación (td) de 14.1 d, con una viabilidad del 80%. Mientras que los cultivos en suspensión suplementados con 1.0 mM de Cr(VI) o Pb(II) mostraron una  y td de 0.0633 d-1 , 10.94 d para el Cr(VI), y 0.0620 d -1 , 11.17 d para el Pb(II), respectivamente. Para evaluar el crecimiento celular y la acumulación de MP se realizaron bioensayos usando concentraciones desde 0.0 a 3.0 mM de Cr(VI) o Pb(II). Las células cultivadas con 1.0 mM de Cr(VI) mostraron un crecimiento relativo (CR) significativamente mayor (122%) que el control (100%); en contraste, las células cultivadas con 2.0 y 3.0 mM de Cr(VI) tuvieron menor CR (81% y 58%, respectivamente) comparado con el control. Por otra parte, las células cultivadas con 1.0 mM de Pb(II) no presentaron diferencias significativas en el CR con respecto al control, mientras que en los tratamientos con 2.0 y 3.0 mM de Pb(II), el CR (81% y 70%, respectivamente) fue significativamente menor que el control (100%). Con relación a la acumulación de MP en las células en suspensión de P. laevigata, se observó una mayor acumulación de Pb (14,086-30,079 mg Pb Kg-1 peso seco (PS)) que Cr (2379-7194 mg Kg-1 PS). Además, se observó que al incrementar la concentración de ambos metales en el medio de cultivo, también se incrementó la acumulación de Cr o Pb en la biomasa. Para determinar la competencia del Cr(VI) por los transportadores de Mn(II) se realizó un bioensayo con Cr(VI)/Mn(II) (0.0, 0.5 y 1.0 mM), en los cuales, se observó que las células presentaron un crecimiento relativo del 83-98% en todas las concentraciones y combinaciones de Cr(VI) y Mn(II). Cuando el Cr(VI) fue adicionado al medio con o sin Mn(II), las células mostraron mayor eficiencia de acumulación para el metal no esencial (Cr) sobre el metal esencial (Mn). Para determinar la competencia del Pb(II) por los transportadores de Zn(II) se realizó un bioensayo con Pb(II)/Zn(II) (0.0, 0.5 y 1.0 mM), en donde se observó que las células presentaron un crecimiento relativo del 63-98% en todas las concentraciones y combinaciones de Pb(II) y Zn(II). Además, cuando el Pb(II) fue adicionado al medio con o sin Zn(II), las células mostraron mayor eficiencia de acumulación para el metal no esencial (Pb) sobre el metal esencial (Zn) (valores del factor de bioconcentración (FBC) para Pb  valores de FBC para zinc; 2-33 veces). Las imágenes de las células observadas con microscopía electrónica de barrido evidenciaron la acumulación del Cr y Pb en la pared celular.
